A 2025 review in Nature Metabolism found that evidence for age-related NAD+ decline in humans varies across studies and tissues, while clinical benefits from NAD+ precursor supplementation remain limited.


NAD+ research is an evolving field, but there is currently insufficient evidence to conclude that NAD+ therapy reverses aging or extends human lifespan.


What Are the Potential Benefits of NAD+ Therapy?

Oral products often contain NAD+ precursors, such as nicotinamide riboside (NR) or nicotinamide mononucleotide (NMN). These compounds are studied for their ability to influence NAD+ metabolism.


Some wellness settings also offer injectable NAD+.


Research on oral precursors cannot automatically be applied to intravenous or intramuscular NAD+ therapy. A 2026 systematic review found that oral NR and NMN can increase certain NAD-related biomarkers, but improvements in meaningful health outcomes have been inconsistent. The review did not identify eligible clinical outcomes trials establishing intravenous or intramuscular NAD+ effectiveness for anti-aging or general wellness.


Is NAD+ Therapy Safe?

Safety depends on the specific formulation, route of administration, product quality and individual medical circumstances.


The FDA has warned about risks associated with improperly compounded injectable NAD+ products, including contamination from ingredients unsuitable for sterile drug preparation. The agency has received reports of severe chills, shaking, vomiting and fatigue following injectable NAD+ use, with some patients requiring medical treatment.
